Spectrophotometric determination of Levetiracetam using p-Chloranilic Acid and Potassium Ferricyanide in pharmaceutical dosage form

Two simple, extractions free spectrophotometric methods (Method 1 and 2) for the quantitative estimation of levetiracetam (LEV) in bulk drugs and pharmaceutical formulations (tablets) have been developed. The first method is based on the charge transfer complexation reaction between drug as n-electron donor and p-chloranilic acid (p-CA) as a π- acceptor to form a violet chromogen measured at 440 nm. The colored product of levetiracetam shows linearity in the concentration range of 50-350 μg/ml. Second method is based on the reaction of levetiracetam with ferric chloride and potassium ferricyanide to form a green colored species having absorption maxima at 750 nm and shows linearity in the concentration range of 100-400 μg/ml. The relative standard deviations for first method is 0.248% and for second method is 0.268% were obtained. Linear relationships with good correlation coefficients (0.998-0.997) were found between absorbance and the corresponding concentrations of the drug. The reliability and performance of the proposed methods was validated statistically the percentage recovery ranged from 99.6 and 99.55 respectively. The results of analysis for the two methods have been validated statistically and by recovery studies.
